In vivo hypermutation and continuous evolution

چکیده
Directed evolution has revolutionized biomolecular engineering by applying cycles of mutation, amplification and selection to genes interest (GOIs). However, classical directed methods that rely on manually staged evolutionary constrain the scale depth search is possible. We describe genetic systems achieve rapid fully inside living cells, enabling continuous GOIs as cells grow. These advance scale, depth, ease overall power access important new areas protein engineering. In vivo a form takes advantage cells. Molina, Rix et al. discuss best practices for designing conducting experiments drug discovery, enzyme fluorescence-activated cell sorting (FACS)-based evolution.

In vivo continuous directed evolution.
The development and application of methods for the laboratory evolution of biomolecules has rapidly progressed over the last few decades. Advancements in continuous microbe culturing and selection design have facilitated the development of new technologies that enable the continuous directed evolution of proteins and nucleic acids. These technologies have the potential to support the extremely ...

Directed evolution remains a powerful, highly generalizable approach for improving the performance of biological systems. However, implementations in eukaryotes rely either on in vitro diversity generation or limited mutational capacities. Here we synthetically optimize the retrotransposon Ty1 to enable in vivo generation of mutant libraries up to 1.6 × 107 l-1 per round, which is the highest o...
متن کاملDirected evolution of mammalian anti-apoptosis proteins by somatic hypermutation.
Recently, researchers have created novel fluorescent proteins by harnessing the somatic hypermutation ability of B cells. In this study, we examined if this approach could be used to evolve a non-fluorescent protein, namely the anti-apoptosis protein Bcl-x(L), using the Ramos B-cell line. After demonstrating that Ramos cells were capable of mutating a heterologous bcl-x(L) transgene, the cells ...
متن کاملEvolution of new nonantibody proteins via iterative somatic hypermutation.
B lymphocytes use somatic hypermutation (SHM) to optimize immunoglobulins. Although SHM can rescue single point mutations deliberately introduced into nonimmunoglobulin genes, such experiments do not show whether SHM can efficiently evolve challenging novel phenotypes requiring multiple unforeseeable mutations in nonantibody proteins. We have now iterated SHM over 23 rounds of fluorescence-acti...
متن کاملContinuous ex vivo and in vivo monitoring with chemical sensors.
The chemical species of principal clinical interest for continuous monitoring include oxygen, carbon dioxide, pH, potassium, and glucose. Although sensors exist for all of these analytes, relatively few continuous monitors are used for blood chemistries. Reviewing the available electrochemical and optical approaches along with the expected system performance requirements provides some insight i...
